Pigs with expression of transgenes may be generated by sperm mediated gene transfer (SMGT).

The SMGT technique consists of the following steps (see schematic presentation here):

• Collection of sperm cells from many different boars
• Evaluation of sperm cell quality: motility, survival rate, morphology and ability to take up exogenous DNA
• Preparation of transgene – linearized DNA is used for uptake in sperm cells
• Removal of seminal fluid and mix with transgene – DNA uptake
• Transfer of transgenic sperm cells to recipient sow by artificial insemination
• Birth and molecular test of transgenic offspring
